Formulation, Optimization and in vitro Evaluation of Apremilast Nanoemulgel for Topical Delivery

Background: Psoriasis is an autoimmune-mediated disease characterized marked by thickened plaques and erythematous skin, which can be treated topical delivery of apremilast. The objective this study was to prepare hydrogel-based nanoemulsions apremilast for psoriasis therapy with a view improving its solubility permeability. Methods: Nanoemulsion components were chosen based on their solubility. Spontaneous emulsification technique used formulate loaded D-optimal design optimize the formulations. optimized formulation assessed globule size, Polydispersity Index, percentage transmittance, entrapment efficiency, pH, Transmission Electron Microscopy nanoemulsion converted into gels using carbopol 940 evaluated critical parameters like spreadability, viscosity extrudability. In vitro drug release ex vivo permeation studies conducted understand kinetics extent permeation. Results: Particle size found in range 141 nm- 245nm. electron microscopic images optimised showed spherical particles nano range. Optimised exhibited excellent efficiency 86% thermodynamically stable. nanoemulgel Franz diffusion cell apparatus appeared follow zero-order kinetics. Ex through porcine skin substantial improvement flux, permeability coefficient, deposition case compared dispersion. Conclusion: results obtained from demonstrated immense potential developed formulations application psoriasis.
